Item Effects of Irisin Compared with Exercise on Specific Metabolic and Obesity Parameters in Female Mice with Obesity(MARY ANN LIEBERT, INC, 2020-01-01) Ozgor, Bahar Yuksel; Demiral, Irem; Zeybek, Umit; Celik, Faruk; Buyru, Faruk; Yeh, John; Bastu, ErcanObjective: We aimed to analyze the changes in metabolic parameters after administration of irisin to obese female mice. Materials and Methods: Sixty mice aged 5-6 weeks were randomized into three groups as irisin, exercise, and control. The control and irisin group remained sedentary, whereas the exercise group started free wheel exercising 6 weeks after the start of the study. The irisin group received irisin after 20 weeks. All mice were sacrificed at the 22nd week of the study, and obesity-related metabolic parameters were analyzed. Results: There was no significant difference between the irisin and exercise groups in weight gain (P > 0.05). By contrast, weight gain in the control group was significantly higher compared with the irisin and exercise groups (P < 0.05). Serum bone morphogenetic protein (BMP), ghrelin, insulin, kisspeptin, leptin, and visfatin levels were statistically lower in the irisin and exercise groups compared with the control group, but no significance was detected between the irisin and exercise groups (P < 0.05 for all parameters). Conclusion: Similar to the effect of exercise, irisin injections resulted in the amelioration of certain obesity-related parameters such as the concentration of adipokines, BMP4, insulin, and ghrelin. Data about the role of ghrelin and ghrelin polymorphisms in the development of acne vulgaris in post-adolescent male patients are limited. Aim: To evaluate the role of serum androgens, insulin resistance, ghrelin and ghrelin polymorphisms in severe acne vulgaris. Material and methods: Thirty-five post-adolescent male patients with a mean age of 28.0 +/- 5.4 years and 33 age-and BMI-matched controls were enrolled. Serum androgens, lipids, insulin sensitivity parameters and ghrelin levels were determined. The PCR method was used for GHRL polymorphisms (rs27647, rs696217 and rs34911341 genotypes). Results: Patients had similar anthropometric measures to controls, except a significantly higher WHR in patients (0.92 +/- 0.06 vs. 0.86 +/- 0.08, p < 0.05). Also, FPG, HOMA-IR values, lipid profile and serum androgen levels were similar. Interestingly, patients had significantly lower ghrelin levels than controls (4.5 +/- 5.8 vs. 101.2 +/- 86.5 pg/ml, p < 0.001). The frequencies of rs696217 and rs34911341 genotypes were similar whereas the distribution of rs27647 alleles was significantly different between the groups (p < 0.05). Current evidence shows that regulatory proteins secreted from the adipose tissue called adipokines may have a role in polycystic ovary syndrome. We planned to investigate the role of endotrophin that has never been researched in polycystic ovary syndrome before and its correlation with other metabolic parameters and adipokines such as adiponectin and ghrelin in patients with polycystic ovary syndrome. Methods: Forty-three women (n: 43) with polycystic ovary syndrome and 43 (n: 43) women as a control group were enrolled in this cross-sectional study. Serum levels of endotrophin, adiponectin, and ghrelin levels were measured with the enzyme-linked immunosorbent assay method. High-density lipoprotein cholesterol, low-density lipoprotein cholesterol, total cholesterol levels, luteinizing hormone/follicle-stimulating hormone ratio, total testosterone, and triglyceride levels were measured. Homeostasis model assessment for insulin resistance index, body mass index, Ferriman Gallwey Score, and waist-to-hip ratio were also evaluated. Results: Total testosterone, homeostasis model assessment for insulin resistance, C-reactive protein, luteinizing hormone/follicle-stimulating hormone ratio, and triglyceride levels were higher in patients with polycystic ovary syndrome (p < 0.01). No difference was detected between the groups in terms of body mass index, Ferriman Gallwey Score, waist-to-hip ratio, total cholesterol, low-density lipoprotein, and high-density lipoprotein levels (p > 0.05). We did not observe any significant difference in adiponectin and ghrelin levels between the groups (p > 0.05). Patients with polycystic ovary syndrome had significantly higher endotrophin levels (p < 0.01). According to our regression analyses {[}area under the curve: 0.973 (0.935-1.000), 95\% confidence interval, 95.2\% sensitivity, and 100\% specificity], it was shown that endotrophin greater than 92 ng/ml and homeostasis model assessment for insulin resistance greater than 2.5 might be good predictors for polycystic ovary syndrome diagnosis. Conclusion: We demonstrated that endotrophin level is higher in patients with polycystic ovary syndrome and may have predicted polycystic ovary syndrome with increased homeostasis model assessment for insulin resistance index. There was no significant difference in adiponectin and ghrelin levels in the polycystic ovary syndrome group. Endotrophin may have a role in polycystic ovary syndrome etiology rather than other adipokines.Item Evaluation of ghrelin and leptin in chronic obstructive pulmonary disease(WOLTERS KLUWER MEDKNOW PUBLICATIONS, 2019-01-01) Uysal, Pelin; Uzun, HafizeOBJECTIVE: Recent studies suggest an important role for ghrelin and leptin in pathogenesis of inflammatory respiratory diseases. Therefore, the present study aimed to investigate ghrelin and leptin levels in the circulation that might be associated with the development of obstruction. METHODS: Stable chronic obstructive pulmonary disease (COPD) patients (n= 119) and matched healthy controls (n = 27) were recruited. The COPD patients were classified into four groups (A, B, C, and D) according to the Revised Global Initiative for Chronic Obstructive Lung Disease guidelines. RESULTS: Compared to controls, significantly decreased leptin and ghrelin levels were observed in all COPD patient groups. Plasma ghrelin concentration was higher in Group A (P < 0.001), Group B (P < 0.001), and Group C (P < 0.05) when compared with the Group D. Plasma leptin concentration was significantly decreased in Group D patients when compared with Group A (P < 0.001) and Group B (P< 0.001).
