Item The Diagnostic Value of Protein Carbonyl Levels in Acute Carbon Monoxide Intoxication(DUZCE UNIV, 2017-01-01) Gunaydin, Mucahit; Turkmen, Suha; Karaca, Yunus; Tatli, Ozgur; Yildirim, Furkan; Altinkaynak, Buket Akcan; Gunduz, AbdulkadirObjective: Carbon monoxide (CO) is the main cause of intoxication-related mortality and morbidity in developed countries. It is responsible for more than half of fatal intoxications in many countries. The purpose of this study was to determine the diagnostic value of protein carbonyl (PC), a good marker of oxidative stress, in association with oxidative stress resulting from hypoxia emerging in patients with acute CO intoxication. Methods: Thirty-four patients diagnosed with acute CO intoxication at the Emergency Department and 38 healthy volunteers were included in the study. Patients' PC levels at time of admission and after treatment were compared with those of a control group. Results: No statistically significant difference was observed among PC levels at time of admission in the patient and control groups (p = 0.305, patient group 0.025 +/- 0.01, control group 0.026 +/- 0.01). A significant decrease was determined in post-treatment PC levels in the patient group compared to those at time of admission (p = 0.006, admission 0.025 +/- 0.01, post-treatment 0.017 +/- 0.008). No significant correlation was determined between patients' carboxyhemoglobin (CO-Hb) levels and PC levels at time of admission (Correlation coefficient = -0.006, p= 0.971). Conclusions: We think that PC is not suitable for use as a biomarker in the acute period in patients with CO intoxication.Item Superoxide Anion Production by the Spermatozoa of Men with Varicocele: Relationship with Varicocele Grade and Semen Parameters(KOREAN SOC SEXUAL MEDICINE \& ANDROLOGY, 2018-01-01) Alkan, Ilter; Yuksel, Meral; Canat, Halil Lutfi; Atalay, Hasan Anil; Can, Osman; Ozveri, Hakan; Basar, Mehmet MuradPurpose: To investigate the pathophysiological role of superoxide anion and total reactive oxygen species (ROS) production by the spermatozoa of men with varicocele and its relationship with varicocele grade and semen parameters. Materials and Methods: This prospective study included 34 men with grade II-III varicocele, regardless of their fertility status. The control group consisted of 13 healthy men. Semen characteristics were examined according to the 2010 World Health Organization criteria. The swim-up method was used for sperm preparation. Total ROS and superoxide anion production was assayed by luminol-and lucigenin-dependent chemiluminescence (CL), respectively. Results: The men with varicocele had significantly higher total ROS and superoxide anion levels than the healthy control subjects (2.9 +/- 0.4 relative light unit (RLU) vs. 2.4 +/- 0.1 RLU, p=0.001 for luminol-dependent CL and 2.8 +/- 0.4 RLU vs. 2.3 +/- 0.2 RLU, p=0.002 for lucigenin-dependent CL). Cases of grade III varicocele had significantly higher superoxide anion and total ROS levels than grade II cases and control subjects (p<0.001). Superoxide anion and total ROS levels were negatively correlated with all semen parameters. Conclusions: The superoxide anion levels produced by spermatozoa were significantly higher in varicocele patients than in control subjects. ROS production was related to increased varicocele grade, impaired semen concentration, and abnormal morphology in men with varicocele. Our findings suggest that superoxide anion overproduction may be an important step in the cascade of ROS-related damage to spermatozoa, resulting in impaired semen parameters in patients with varicocele.Item XPD and hOGG1 gene polymorphisms in reperfusion oxidative stress(FUNPEC-EDITORA, 2011-01-01) Isbir, O.; Gormus, U.; Ergen, H. Human 8-oxoguanine glycosylase 1 (hOGG1) is an enzyme to repair specific DNA lesions and a good marker of hydroxyl radical damage to DNA. XPD is another DNA repair gene. Polymorphism analyses were done by PCR-RFLPItem Effects of Alprazolam and Melatonin Used for Premedication on Oxidative Stress, Glicocalyx Integrity and Neurocognitive Functions(AVES, 2018-01-01) Can, Meltem Guner; Ulugol, Halim; Gunes, Isin; Aksu, Ugur; Tosun, Melis; Karduz, Gulsum; Vardar, Kubra; Toraman, FevziObjective: The purpose of this investigation was to compare the effect of alprazolam and melatonin on oxidative stress, glicocalyx integrity and neurocognitive function in patients undergoing coronary artery bypass grafting (CABG). Methods: Overall, 42 patients undergoing CABG were retrospectively included in this study. Blood samples which preserved at -70 degrees C for a previous study were used for this study. The participants were divided into two groups. Patients in the Group A were administered alprazolam before the operation, whereas melatonin was used for premedication in the Group M. Blood samples were collected at three time points {[}T0: before anaesthesia induction, T1: admittance to intensive care unit (ICU), T2: 24 h after ICU admission], and oxidative stress parameters and glicocalyx integrity were evaluated. Furthermore, Mini-Mental State Examination was recorded to measure neurocognitive function. Results: The total thiol levels which were measured as an antioxidant parameter were significantly higher, and free Hb values were significantly lower in the Group M compared to the Group A (p<0.05). No significant differences were found in order to oxidative stress parameter levels, extubation time, length of hospital stay, durations of cross-clamp, cardiopulmonary bypass and operation and Mini-Mental State Examination results between the two groups (p>0.05). Conclusion: In light of positive effects on oxidatif stress parameters, melatonin may be considered as a good and safe premedication agent with its anxiolytic, antioxidant and minimal haemodynamic and respiratory effects.
